Through its inhibitory action on CYP3A4, nefazodone enhances the actions of several drugs including carbamazepine, clozapine, HMG-CoA reductase inhibitors (“statins”), and TCAs. Though rare, nefazodone has caused life-threatening hepatotoxicity requiring liver transplantation.
